Why we included this project

Anyone working with high-throughput RNA-seq data who needs to tell vaccine-derived reads apart from genuine SARS-CoV-2 signal will find this useful. The project publishes experimentally assembled spike-encoding mRNA sequences recovered from leftover Moderna and Pfizer/BioNTech vaccine vials, including a working assembly of the Moderna construct whose exact sequence was not publicly documented at the time of the work, and a confirmation of the previously reported Pfizer/BioNTech sequence. Clinical and research teams can use these sequences to recognize therapeutic mRNA in their samples and avoid misclassifying it as host or viral material, whether they are doing variant surveillance or designing PCR and LAMP tests. The notes also walk through the actual wet-lab and sequencing workflow, from extraction to template-switch amplification and strandedness analysis, which is helpful for labs trying to reproduce the approach. It is a scholarly dataset and protocol rather than a software tool, so treat it as a reference resource instead of something to install and run.
